It should be the "State Championship Division" as the top 10 teams in Big V, then the remaining teams all in "State League One" split into East/West conferences, playing only within that conference for the league title.
Then both conference winners play each other for the overall league Championship and for promotion, neutral venue at the SBC.

Melbourne Boy - Youth Champs run two pools then with cross overs for finals. Seems like a good system that your State League 1 concept could use. That would work well I reckon. Also reduce travel for the regional teams e.g. Pakenham to Mildura

Pakenham to Mildura is the same as anyone to Mildura from Melbourne, they mostly all fly out from the airport.

Makes a lot of sense the way you pitch it, I'd be very much in favour of that.
Most of these programs run at a loss anyway, so why not minimise some of the travel as much as possible and keep them in regional conferences.
Maybe a Final 4 to determine top-4 placement (assuming they'll have to play in the grading tournament the year after?)

Grading tournaments are a good idea for YL, but no need in Seniors.
State League One winner gets promoted to Championship, and the Champ division Wooden Spoon gets relegated to League One, even have the bottom two teams playoff for survival first week of the finals.
